Abstract
This AOP describes the relationships between an increase in bulky DNA adducts, inadequate repair, and an increase in mutations. Bulky DNA adducts are formed when activated genotoxic aromatic compounds interact with the nitrogenous bases of DNA. While not all bulky DNA adducts are stable, persistent adducts can be inadequately repaired due to incorrect repair if the adduct favors incorporation of an incorrect base opposite the lesion. Inadequate repair of bulky DNA adducts can also occur due to lack of damage repair or due to incomplete repair. Inadequate repair (due to incorrect repair, lack of damage repair or incomplete repair) can then lead to the formation of mutations during later replication.
